I'm a high school biology teacher trying to make our genetics unit more hands-on, and I want to do the classic DNA extraction from fruit experiment with my students, but I'm looking for ways to elevate it beyond just a simple demonstration. I've used strawberries with dish soap and alcohol before, but I want the students to be able to compare yields or purity between different fruits or extraction methods, perhaps even quantifying the DNA somehow. For educators or hobbyists who have done this, what are the most reliable and engaging protocols for a classroom setting that yield visible, spoolable DNA? Have you successfully incorporated variables like different salt concentrations, types of alcohol, or mechanical disruption techniques to turn it into a genuine inquiry-based lab, and are there any safe stains or simple spectrophotometry methods accessible for high school labs to analyze the results?
